Öğe Bisphenol A levels in bowel endometrioma diagnosed serums: A case control study(2022) Ardiç, Filiz; Çelik, Humeyra; Yeşilyurt, Hüseyin; Otçu, Serap Mutlu ÖzçelikAim: To investigate the bisphenol A (BPA) levels, which may be a risk factor in the etiology of\rendometrioma, in patients diagnosed laparoscopically with endometrioma with and without bowel\rinvolvement.\rMethod: In the prospective cross-sectional case control study, 47 cases were included in the study, which were\radmitted to the gynecology and infertility services with and without bowel involvement endometrioma who\rwere operated and diagnosed histopathologically. 43 patients were included in the control group. For serum\rBPA value, blood samples taken immediately before the operation were studied in laboratory. Patients and\rcontrols were compared with controls in terms of serum BPA values.\rResults: The mean age of the patients was 35 ± 2 in the endometriosis group and 36 ± 2 in the control group\rwhich was and not statistically significant. There was no statistical difference between the patient and control\rgroups in terms of menstruation periods. Serum BPA levels were significantly higher in the bowel involvement\rgroup compared to the non-bowel involvement group, as the distribution width was higher due to excessive\rvalues, and only 5 patients with bowel involvement did not reach statistically significant levels. Serum BPA\rlevel was 1084±1132 ng/L in the endometriosis group and 269±99 ng/L in the control group which was\rstatistically significant (p<0,001).\rConclusions: BPA levels were showing very wide range especially in the patient group. Serum BPA levels\rwas statistically significantly higher in the endometrioma group compared to the control group. Therefore, in\rthe etiology of endometriosis BPA may take a definite place.Öğe Cytotoxic effects of calcitriol on osteosarcoma cells(2022) Çelik, Humeyra; Çetinkaya, Ayhan; Celik, IlhanIntroduction: It was aimed to examine the effect of calcitriol which has known anticancer activities, on the osteosarcoma cell line SAOS-2 cells [1,25(OH)2D3]. Methods: SAOS-2 cells were grown in culture in conventional culture flasks in DMEM medium at 37°C and 5% CO2. When the cells were 70-80% confluent, morphological changes were examined under an inverted microscope. The cells were passaged into 96 microplates, and after passage, different concentrations of calcitriol was applied to the cells (0.1; 0.5; 1; 5; 10; 25 nM/ml) was done. After administration, cytotoxic effect and proliferation rates/cell proliferation were analyzed by MTT method. Results: The effect of calcitriol applied at different concentrations in cultured cells were 0.1, 1, 10, 25 nM/ml doses, in these groups proliferation was found to be statistically significantly reduced compared to the control group (p?0.05). Conclusion: Our data imply that the antiproliferative effects of vitamin D applications can be benefited from in exosomal, combined and supportive treatments in osteosarcoma. Further trials are warranted to confirm and validate our findings.Öğe The Effect of Wet Cupping Therapy on Anxiety and Thiol/Disulfide Homeostasis in Premature Ovarian Failure: An Experimental Study(2022) Çelik, Humeyra; Çetinkaya, Ayhan; Bozat, Bihter Gökçe; Düzcü, Selma Erdoğan; Alişik, Murat; Yis, Özgür MehmetObjective: The effect of wet cupping therapy on anxiety\rand thiol/disulfide homeostasis was evaluated in rats with premature\rovarian failure (POF) induced by cyclophosphamide (CYC). Material\rand Methods: Twenty four Wistar Albino female rats were divided\rinto control (n=6), POF (n=6), wet cupping (n=6), POF+wet cupping\r(n=6) groups. CYC at a dose of 200 mg/kg was injected intraperitoneally\rto POF and POF+wet cupping groups. One week later, wet cupping\rtherapy was applied to wet cupping and POF+wet cupping groups for\r5 minutes. The next day, Open Field Test and Elevated Plus Maze Test\rwere applied to all experimental groups to evaluate anxiety. After sacrification,\rto assess pattern formation, endometrial thickness and ovarian\rreserve were measured with hematoxylin-eosin staining.\rThiol/disulfide homeostasis and erythrocyte oxidized-reduced glutathione\rlevels were measured in serum to measure oxidative stress. Results:\rAlthough there was no statistical significance, it was observed\rthat anxiety decreased in POF+wet cupping compared to POF group.\rThere was no change in thiol-disulfide values in POF+wet cupping\rcompared to POF group (p>0.05). Conclusion: Wet cupping therapy\rdid not reduce anxiety in the experimental POF model and did not shift\rthe balance to positive in thiol/disulfide homeostasis in favor of thiols.\rHowever, the obtained graphics, numerical data and high standard deviation\rsuggest that meaningful data could be obtained with more subjects.Öğe Histopathological effects on kidney of diclofenac potassium and diazepam used in an experimental epilepsy model(2023) Saylan, Aslihan; Turel, Canan Akünal; Türel, Idris; Çetinkaya, Ayhan; Torun, Ibrahim Ethem; Çelik, HumeyraAim: To investigate the effects of diazepam, which has anticonvulsant and anxiolytic effects, and diclofenac potassium, which has anti-inflammatory, analgesic and antipyretic effects, on rat kidney tissue, used in an experimental epilepsy model. Methods: 32 Wistar albino rats (2-4 months old, 200-250 gr) were used in the study. The rats were grouped in four as 8 rats in each group: Epilepsy, Epilepsy + Diazepam, Epilepsy + Diclofenac potassium, Epilepsy + Diazepam + Diclofenac potassium. Epileptic seizure model was created with penicillin (500.000 IU) injected intracortically under urethane anesthesia. 30 minutes later, diazepam (0.1 mg/kg) and diclofenac potassium (10 mg/kg) were administered intraperitoneally. At the end of the study, rat kidneys were removed and evaluated histopathologically in terms of inflammation, glomerular shrinkage, tubular dilatation, tubular epithelial thinning, desquame epithelium, brush epithelial loss, vacuolization, hemorrhage and congestion. Results: No difference was found between diazepam and diclofenac potassium in terms of vacuolization, glomerular shrinkage, tubular dilatation and hemorrhage. Inflammation, congestion and tubular epithelial thinning rate were found to be lower inEpilepsy + Diclofenac potassium and Epilepsy + Diazepam + Diclofenac potassium group when compared with Epilepsy + Diazepam group. While brush epithelial loss and desquame epithelial rate was found to be lowest in the epilepsy group, these parameters were not found to show a significant difference between drug groups. Conclusion: It was concluded that combined use of diazepam and diclofenac potassium in their effects on kidney are more useful than their single use.
