In Vitro Şartlarında Küf Etmenlerine Karşı Tavuk Gübresinden Elde Edilmiş Odun Sirkesinin Antifungal Etkisi

Özet
Bu çalışma, Broyler tavuk yetiştiriciliği atığından elde edilen odun sirkesi’nin, Aspergillus niger ve Penicillium digitatum mikrofungusları üzerinde antifungal etkisini saptamak amacıyla in vitro şartlarında yürütülmüştür. %1, %3, %5, %7 ve %10 mL'lik konsantrasyonlarında odun sirkesi içeren PDA besiyerlerine, patojen kültürlerin 5 mm çapında miselyum diskleri ekilmiş ve 24±1 oC’de 7 gün inkübasyona bırakılmıştır. Uygulama, Tesadüf Parselleri Deneme Desenine göre 3 tekerrürlü olarak yürütülmüştür. İnkübasyon süresi sonunda, fungusların koloni çapları ölçülmüş ve kontrole göre odun sirkesinin % engelleme oranları hesaplanmıştır. Yapılan uygulamada, Tür x İlaç dozu interaksiyonunun (P=0.000) ve aynı doz odun sirkesinde, A. niger ile P. digitatum’un misel gelişimleri arasındaki farkların önemli olduğu (P?0.05) ve %10 mL’lik doz uygulamasının P. digitatum’a karşı fungisidal etki gösterdiği tespit edilmiştir. Sonuç olarak; odun sirkesinin, A. niger ve P. digitatum’un misel gelişimlerini engellediği tespit edilmiş olup, hastalık etmenlerine karşı denenmesinin faydalı olabileceği düşünülmektedir.
In this study was conducted to test the antifungal effect of wood vinegar, derived from broiler chicken manure, on Aspergillus niger and Penicillium digitatum microfungi in in vitro conditions.Wood vinegar at doses of 1%, 3%, 5%, 7% and 10% were added in to the PDA growing media. Inoculum sources, taken from the outer margins of growing fungal cultures in 5 mm mycelial discs, were transferred to PDA media. After the inoculation, fungal cultures were kept in incubation at 24 ±1 Cº for 7 days. Experiments were set up in Randomized Plot Design with three replicas. Seven days later, colonial diameters of fungal cultures were measured and the inhibition rates (%) of wood vinegar were calculated compared to control treatment. Results revealed that there was a meaningful difference of applied doses on the species, Species X Ingredient interaction (P=0.000) and same dose of wood vinegar had different effect on mycelial development of A. niger and P. digitatum that were found statistically significant (P?0.05). It was determined that a dose of 10% mL had a fungicidal effect against P. digitatum. As a result; it has been determined that A. niger and P. digitatum inhibit mycelial growth of wood vinegar and it is thought that it may be useful to try against disease agents.
